Development of assays for the capture and detection of prostate cancer-derived exosomes and exosomal markers

In prostate cancer there is a great need for improved biomarkers of disease, ideally multiplex markers with some predictive utility. Prostate cancer-derived nanovesicles (50-150nm) called exosomes, which do not contain proteins secreted by the Golgi secretory pathway (such as PSA), but are enriched in cancer-derived molecules (including prostate cancer markers such as TMPRSS2-ERG fusion mRNAs), are secreted into the circulation or urine, which also contain exosomes from other cells.

This nascent field currently lacks the tools required for selective isolation of exosomes from complex fluids, and those for quantifying prostate cancer-derived exosomes. The molecular repertoire of exosomes and how this varies during prostate cancer progression has not yet been well explored. This global collaborative project will develop novel reagents and assays for exosome isolation and quantitation and cross validate these against other capture techniques in different laboratories. Through analysing broad retrospective and prospective cohorts of prostate cancer-patients, this international collaborative study aims to identify and validate new panels of exosome-related molecules predictive of:

  • indolent vs aggressive disease (localised disease)
  • prognosis for patients with metastatic disease,
  • chemoresponse/resistance for patients with advanced disease who are undergoing therapy.
