This innovative project will develop a new hierarchy of multiscale mathematical models that will be used to quantify how ghrelin modulates cell behaviour (motility, proliferation and death) and provide insight into the precise details of how ghrelin promotes cell invasion. This project will demonstrate the potential for ghrelin-based strategies to control cell invasion.

This project seeks to define cancer survival pathways induced by insulin in prostate cancer cells, in the context of ADT that lead to prostate cancer survival, growth and metastasis. This leads to the logical speculation that there may be therapeutic benefit to using inhibitors of insulin and downstream molecules, currently in clinical use or development, to prevent insulin induced CRPC pathways.

In order to improve capture and molecular and cellular analyses of Circulating Tumour Cells and CTMs in prostate cancer patients, we will compare and refine methodologies via a collaborative approach. As a collective team, we have complementary prostate cancer patients on defined clinical trials or protocols which we will use to assess identified individual biomarkers and/or biological classes or signatures to provide biological information with potential clinical significance to prostate cancer progression and metastasis.

The molecular repertoire of exosomes and how this varies during prostate cancer progression has not yet been well explored. This global collaborative project will develop novel reagents and assays for exosome isolation and quantitation and cross validate these against other capture techniques in different laboratories.

This research will reveal the underpinning mechanism of current advances in advanced prostate cancer clinical trials with the new agent Abiraterone. The innovative research proposed in this study will enable significant advances in understanding the driving force underpinning androgen synthesis in prostate tumours, linking for the first time the potential for high levels of insulin to act directly on prostate tumour cells to stimulate this synthesis.
